Study on polyphenols,antioxidant and anticancer activities of methanol extract from olive leaves

Abstract:

The methanol extract of olive leaves (EOL) was obtained by ultrasonic-assisted method.The polyphenols of EOL were determined by HPLC,and they antioxidant and anti-tumor activities were explored by Caenorhabditis elegans and Hela cells.The results showed that EOL contains 5 main polyphenol components including rutin,luteolin-4′-O-glucoside,apigenin-7-O-glucoside,luteolin and apigenin.EOL significantly increased the antioxidant enzyme activities of SOD,CAT and GSH-Px,decreased the level of MDA,and increased the survival rate of C. elegans under oxidative stress by 72.52%.In addition,HeLa cells were treated with different EOL concentrations to determine the inhibition rate of HeLa cells and the underlying anti-tumor mechanism of EOL was also explored.The results showed that EOL inhibited the proliferation of Hela cells by up-regulating the activities of Caspase 9 and Caspase 3,and the tumor cells inhibition rate was concentration and time-dependent.Therefore,EOL has the potential to be developed as an antioxidant and anticancer drugs.
