Flavonoids of Rhizoma Sparaganii Induce S/G2 Stage Arrest in A549 and MCF-7 Cells

Abstract:

To investigate the cytotoxicity effects and mechanisms on estrogen receptor positive (ER+) cancer cell lines A549 and MCF-7 of the Rhizoma Sparganii flavonoids (RSF),A549 or MCF-7 cells in DMEM mediums with 188,375,and 750 µg/mL RSF were cultured for 3 days,respectively. DMEM with 1% methanol culture cells as control. The cell proliferation activity,anti-a-tublin form,apoptosis as well as cell cycle differences between groups were studied by MTT,immunofluorescence,and flow cytometry technology. As a result,compare to the 1% methanol control groups,RSF could inhibit the proliferation activity,increase the rate of apoptotic bodies,and induce cytoskeleton disorder of the A549 and MCF-7 cells with the dose-related. In addition,S/G2stage arrests were induced obviously in A549 and MCF-7 cells cultured with 375 and 750 µg/mL RSF. The results indicated that RSF showed a notable cytotoxicity against A549 and MCF-7 cells,and the inhibitions of cell proliferation were related with S/G2 cell cycle arrests.
