Optimization of Extraction Technology of Forsythiaside and Phillyrin in Forsythia suspense Leaves Based on Response Surface Methodology

Abstract: Response surface methodology (RSM) was applied to optimize the extraction technology of forsythiaside and phillyrin from forsythia suspense leaves. Base on single factor experiment, the influence factors of liquid to solid ration, ethanol concentration, extraction time and extraction temperature were studied on the yields of forsythiaside and phillyrin. According to Box-Behnken combination method (Design expert 8.0.5 b), the optimal conditions were determined as follows: ratio of liquid to solid 30 mL/g, ethanol concentration 53%, extraction time 53 min, and extraction temperature 74 ℃. Under the conditions, the yields of forsythiaside and phillyrin were 7.56% and 3.09%, respectively, and the relative error between the testing values (7.53% and 3.03%, respectively) and theoretic values were 0.4% and 2.0%, respectively.
