Determination of Glutathione in Agaricus blazei Murriu by HPLC

Abstract: A simple,quick and reliable method was established to determine the content of glutathione (GSH) in Agaricus blazei Murriu by HPLC. The sample was ultrasonically extracted by distilled water. The HPLC analysis conditions were developed and optimized as follows:0.1% trifluoroacetic acid and methanol (98:2,v/v) were used as mobile phases with the flow rate of 1.0 mL/min,The column oven temperature was set at 35 ℃.The detection wavelength was set at 200 nm.The presented method showed good linear relationship in the range of 8 μg/mL-160 μg/mL, R2=0.9997,and the detection limit of GSH was 0.125 μg/mL,the recovery was between 94.18%-95.80% with the RSD less than 2% (n=9).The developed HPLC method was suitable for the analysis and determination of samples in large quantity.
