In vitro Culture of Huperzia serrata Thallus for the Medicinal Component Production

Abstract: In this research, in vitro culture method was developed for obtaining Huperzia serrata thallus which producets Huperzine A,an anti-Alzheimer’s disease (AD) drug candidate.The semi-woody branches of wild mature plant of huperzia serrata from Lushan were chosen to be the test material.The test results indicate that the suitable medium to induce thallus was 6,7-V+ IAA0.5 mg/L,and its differentiation rate was 50%.Also,MS minimal medium without any hormone was established as the optimal subculture medium.Given sugar amount of 20 g/L in this subculture medium, the relative increasing rate of thallus was 1362.6% after 55 days, namely 14.6 folds biomass of inoculation.Through Thin-layer Chromatography and High Performance Liquid Chromatography detections,the production of active ingredients Huperzine A by the obtained thallus has been confirmed.
