Optimization of Ultrasonic-Assisted Extraction of Lentinan by Response Surface Methodology

Abstract: In this study,Response Surface Methodology (RSM) was applied to optimize the extraction of lentinan.On the basis of the previous one-factor test results,ultrasonic power,ultrasonic time,extraction temperature,extraction time were selected as influencing factors and the extraction yield of lentinan was used as the response value in RSM.According to the orthogonal rotation combination experiment design,the method of response with four factors and three levels was adopted.The results showed that the optimal extraction conditions were as followed:in 174.94 W of ultrasonic power,ultrasonic time 18.94 min,in 80.71 ℃ extract 3.01 h,the highest extraction yield reached at 9.61%.When the concentration of lentinan was 3 mg/mL,the ·OH scavenging rate was determined to be 52.1%.
