Optimization of Purification Conditions of Flavonoids from the Stem of Hypericum augustinii N.Robson by Macroporous Resin

Abstract: The optimal macroporous resin for the purification of flavonoids from the stem of Hypericum augustinii N.Robson was screened through static adsorption.The technological parameters of purification process were optimized using static adsorption and desorption kinetics.The results showed that AB8 was the best macroporous resin.The optimal technological parameters were:the concentration of sample solution was 1.30 mg/mL,the sample volume was 60 mL,pH = 4.0 and the flow velocity was 1.00 mL/min.The diameter height ratio of resin column was 1:10,and the elution agents was 70% ethanol (pH = 7.0).The purity of flavonoids from the stem of H.augustinii increased from 30.26% to 55.70% by AB8 macroporous resin.The developed purification process was simple and feasible.
