Extraction and Deproteinization of Polysaccharides from Guizhou wild Paederia scandens

Abstract: A process for extraction of polysaccharides from Guizhou wild Paederia scandens powder was optimized through single factor test and orthogonal test.Four different methods,namely Sevag,TCA,Sevag-TCA,and HClO4 methods,for the removal of protein were investigated.The results showed that HClO4 method was demonstrated to be the best way for the removal of protein,and a protein emergence rate of 94.78% can be achieved after deproteinizing protein for 3 times.Moreover,15.14% polysaccharide loss rate was detected under the optimal extraction conditions of using water as solvent,solid-liquid ratio of 1:50,and ultrasonic power of 70 W at a bath temperature of 70 ℃ within an extraction time of 110 min repeating for two times.This process was simple and low cost.
