Effects of Crocin on Bcl-2, Bax and Caspase-3 Expression of PC12 Cells Injured by H2O2

Abstract: In this study,the effects of crocin on Bcl-2, Bax and Caspase-3 expression of PC12 cells injured by H2O2 were investigated.The possible mechanism of its protective effect on Alzheimer’s disease (AD) model cells was further explored.The effect of crocin on PC12 cells was detected using MTT and LDH assays.The expression of mRNA and protein (Bcl-2, Bax and Caspase-3) were detected by RT-PCR and Western Blot assays.With the concentration of crocin at 0.625 μM and 5 μM,the cell survival rate increasing in a dose-dependent manner.The expression levels of mRNA and protein of Bcl-2 increased,while expressions of Bax and Caspase-3 were decreased in crocin group and VE group (compared with the model group, P<0.05).These results indicated that the antioxidant and antiapoptosis effects of crocin were possible induced through increasing expression of Bcl-2 in PC12 cells.
