Screening for Active Components of Anti-angiogenesis from Chinese Medicine Using Zebrafish Model

Abstract: In order to provide lead compounds for further research and development of anti-tumor drug,fluorescent transgenic zebrafish model was used to screen for components with antiangiogenesis activity fromtraditional Chinese medicine,and to compare their effects on inhibiting angiogenesis,VEGFR2:GFP transgenic zebrafish with fluorescent blood vessels was taken as model,and zebrafish embryos were treated with 18 components from different Chinese medicine with the concentration of 25,50 and 100 μg/mL,respectively.The effects of these components were observed and quantitatively analyzed.The results showed that 18 different compositions had effects on inhibiting angiogenesis in different degrees.Those components from Chinese medicine with cold or hot natures all had significant effect,while the other ones with neutral nature have little effect.Based on the theory of nature of Chinese medicine,the effects of the active components on inhibiting angiogenesis were analyzed,and five components were screened for developing new anti-cancer drug,including tetrandrine hormone,akeboside D,ferulic acid,tanshinone IIA and catechin.This study provided a new insight into screening lead components for developing new drug with anti-cancer activity based on the theory of traditional Chinese medicine.
