Estimation of Acute and Genetic Toxicity of Polyprenols from Ginkgo biloaba L.

Abstract: In this study,standardized acute toxicity test in mice and Ames test,bone marrow polychromatie erythrocyte micronucleus test and primary spermatocyte chromosome aberration tests in rats were conducted to evaluatre the acute and genetic toxicity of polyprenols of Ginkgo biloba L (GBP),a new type of lipids with isoprenyl units.The results showed that GBP was nontoxic,the maximum drug dose was 21.5 mg/kg in mice.No difference was detected between different groups on the body weight,growth organ/body weight ratio of rats and biochemical indexes (P>0.05).When GBP dose was 5000 μg/dish with or without S9,spontaneous revertant colonies of TA97,TA98,TA100 and TA102 were within the normal range,and no significant mutagenic activity were detected in Ames assay (P>0.05). Compared with negative control group in the micronucleus test,when GBP dose was 10 g/kg b·wt,the ratio of polychromatic erythrocytes to positive red cells in rats was not statistically significant (P>0.05),and proliferation of bone marrow erythrocytes system had no obvious inhibition,and the observation of polychromatie erythrocyte micronucleus had no obvious effect.Compared with negative control group in the chromosome aberration test,the aberrations rate of primary spermatocytes chromosome was no statistical difference (P>0.05),and no significant mutagenic activity of primary spermatocytes chromosome for male mouse were detected.Therefore,GBP was non-toxic,non-carcinogenic,non-teratogenic and non-mutagenic.
