Simultaneous Determination of 6 Nucleoside Components in Corydalis yanhusuo W.T.Wang by High Performance Liquid Chromatography

Abstract: To establish a HPLC method for the simultaneous determination of 6 nucleoside components in Corydalis yanhusuo W.T.Wang.The assay was performed on a Grace Prevail C18 (4.6 mm×250 mm,5 μm) column;mobile phase consisted of methanol (A)-0.1% orthophosphoric acid water (B) with gradient elution at the flow rate of 0.8 mL/min;column temperature was 30 ℃;and the detection wavelength was set at 260 nm.The investigated compounds,including cytidine,uridine, adenosine, 2-deoxyadenosine,guanosine and thymidine,showed good linearity (r≥0.9994) in the range of 6.70-134.06, 9.08-181.68, 9.68-193.52, 2.11-42.12, 6.80-135.94 and 2.56-51.26 μg/mL,respectively.The average recoveries (n=6) were within 99.7%-102.1% with RSD≤2.3%.The contents of 6 nucleoside components in C.yanhusuo samples from Nantong city were:20.97,207.10,221.01,6.82,152.52 and 11.60 μg/g;in C.yanhusuo samples from Hangzhou city were:21.58,207.69,220.70,8.37,154.20 and 11.52 μg/g and in C.yanhusuo samples from Yixing city were: 20.49,208.62,220.93,8.59,157.27 and 11.38 μg/g.The developed HPLC method was simple and valid.It can be used for quality evaluation and quality control of C.yanhusuo.
