Extraction and Monosaccharide Composition of Andrias davidianus skin mucus polysaccharide

Abstract: In this study,the extraction and monosaccharide composition of polysaccharides from skin mucous of Andrias davidianus were studied.Using water extraction, alkali extraction, acid extraction and enzyme extraction,crude polysaccharides were extracted from skin mucous of A.davidianus. The components of polysaccharides were determined by 1-phenyl-3-methyl-5-pyrazolone (PMP) pre-column derivatization-HPLC method.Results showed that carbohydrate content of alkali extract,acid extract,water extract,alkaline protease extract and acid protease extract was 4.23%,4.23%,1.81%,3.71%,2.19% respectively.The monosaccharide composition of alkali extract was mannose (Man),glucuronic acid (GlcUA),galacturonic acid (GalUA),glucosamine (GlcN),glucose (Glc) and galactose (Gal);the monosaccharide composition of acid extract was Man,GlcUA;water extract was Man,GlcUA and Glc;the monosaccharide composition of alkaline protease extract was Man,GlcUA and GlcN;acid protease extract was Man,GlcUA and Glc.This study provided basis for the development and utilization of A.davidianus skin mucus polysaccharides.
