Protective Effect of Schisandra chinensis Fructus on Hydrogen Peroxide-induced Oxidative Stress Injury in HaCaT Cells

Abstract: In this study,the protective effects of different fractions of Schisandra chinensis Fructus on H2O2-induced oxidative stress injury in HaCaT cells were investigated and the active components were screened.The extracts of S.chinensis fruits were separated with macroporous resin.With DPPH free radical scavenging activity and total antioxidant capacity as indexes,the antioxidant activities of S.chinensis were assessed.MTT method was used to screen the protective effects of different fractions of S.chinensis on H2O2-induced oxidative stress injury in HaCaT cells.The active components with better protective effect were obtained.The results showed that 50% EtOH fraction demonstrated the strongest antioxidant activity and can inhibit the H2O2-induced injury in HaCaT cells.Furthermore,50% EtOH fraction reduced MDA activity and elevated SOD,GSH activities.The results suggested that 50% EtOH fraction was the active components of S.chinensis protecting skin cells against H2O2-induced oxidative stress injury.
