Simultaneous Determination of 7 Isoquinoline Alkaloids from Corydalis yanhusuo W.T.Wang by High Performance Liquid Chromatography

Abstract: In this paper,a HPLC method was established for the simultaneous determination of palmatine, berberine, dehydrocorydaline, tetrahydropalmatine, isocorybulbine, corydaline and tetrahydrocoptisine in Corydalis yanhusuo W.T.Wang,and to investigate content of 7 isoquinoline alkaloids in C.yanhusuo samples from different resources.The chromatographic separation was carried out on an Agilent SB C18 column (4.6 × 250 mm,5 μm) with acetonitrile-0.1% acetic acid aqueous solution (adjusted pH to 5.0 by triethylamine) as mobile phases,flow rate was 1.0 mL/min,the detection wavelength was set at 280 nm.Good linearities of palmatine, berberine, dehydrocorydaline, tetrahydropalmatine, isocorybulbine, corydaline and tetrahydrocoptisine were achieved within the range of 2.0-40.1, 2.0-39.5, 5.1-101.3, 5.0-99.8, 2.1-41.2, 5.0-100.1 and 2.0-39.7 μg/mL,respectively.The average recoveries were 95.6%,96.1%,96.5%,101.4%,101.9%,97.3% and 102.3%;The RSD were 2.77%,2.50%,3.33%,4.18%,2.93%,2.86% and 2.60%,respectively.The content of 7 isoquinoline alkaloids in C.yanhusuo samples was distinct difference from different resources.The developed method was accurate and can be used for quality control of C.yanhusuo.
