Immunoregulation Effect of Chlorogenic Acid on Mouse Peritoneal Macrophages

Abstract: This study was aimed to explore the immunoregulation effects of chlorogenic acid (CGA) on normal and LPS-stimulated mouse peritoneal macrophages (PM).MTT assay was used to test the proliferation of macrophage.The phagocytic activity of PM was tested by neutral red phagocytosis experiments.Moreover,the production of nitrous oxide (NO) was measured by Griess reaction and the production of interleukin-1β (IL-1β),tumor necrosis factor-α (TNF-α) and interleukin-10 (IL-10) in culture supernatants were measured by Enzyme-Linked Immunosorbent method (ELISA).The results showed that CGA promoted the proliferation and the phagocytic activities,increased the secretion of NO and proinflammatory cytokine (IL-1β,TNF-α),inhibited the secretion of anti-inflammatory cytokines (IL-10) both in normal macrophages and in LPS-activated macrophages in a dose-dependent manner.
