Evaluation of the quality of Nymphaeae Flos from different origins based on fingerprint combined with chemical pattern recognition

Abstract:

HPLC fingerprint of Nymphaeae Flos was established,the content of five components was determined,and the composition differences of Nymphaeae Flos in different origins were comprehensively analyzed by combining with chemical pattern recognition method,which laid the foundation for further development and utilization of Nymphaeae Flos.The HPLC fingerprint was established by Waters C18 (250 mm×4.6 mm,5 μm) column.The mobile phase was acetonitrile-0.2% phosphoric acid aqueous solution.The flow rate was 1.0 mL/min.The column temperature was 30 ℃.The detection wavelength were 273 nm,350 nm.At 273 nm,23 common peaks were identified and five chemical components were identified.At 350 nm,17 common peaks were identified and four chemical components were identified.In the cluster analysis,15 batches of samples were divided into five groups at 273 nm and three groups at 350 nm.Orthogonal partial least squares discriminant analysis (OPLS-DA) identified 10 different markers at 273 nm that could affect the quality of Nymphaeae Flos.The similarity of 15 batches of Nymphaeae Flos samples was 0.972 to 0.999,indicating that different batches of Nymphaeae Flos samples were different.This method can effectively evaluate the quality difference of Nymphaeae Flos from different habitats and provide reference for its quality control.
